Materials
EMM -all
フタル酸水素カリウム 3 g
Na2HPO4 2.2 g (注意：NaH2PO4と間違えないように)
50x Salts 20 ml （Working 4℃, stock -20℃）
1000x Vitamins 1 ml （Stock at 4℃）
10000x Minerals 100 ul (Working 4℃, stock -20℃)
(Agar) if you want to make plates, 20 g Agar is added.
DW 800 ml
total ~820 ml
--> autoclave
--> after autoclave, supplements are added. (or store at 4℃ w/o adding supplements)
supplements
20% Glucose (10x) 100 ml C (final 2 %)
20% NH4Cl (50x) 20 ml High N
final 20 mM Glutamate High N
final 20 mM Phenylalanine Low N (suit for G1/S checkpoint analysis)
final 20 mM Isoleucine Low N
(Aminoacids for N source are added as powder. Medium is then filter-sterilized.)
10 mg/ml Thiamine 500 ul
Adenine, Leucine, Uracil, Histidine, Lysine as you need.

x10 Glucose 5 ml
x50 NH4Cl 1 ml
DDW 3 ml
total 50 ml
Remarks
It is important for reducing background to add supplements after autoclave. If you want to reduce the background for imaging, media can be sterilized by filter instead of autoclave.
